CGI-58 is a membrane protein that is essential for plant growth, development and lipid metabolism. We have shown previously that expression of mouse Order CGI-58 antibody  cells increases phosphatidylglycerol (PG) and decreases other major phospholipids including PC, PE, phosphatidylinositol, and phosphatidylserine. Interestingly, a mutant of plant CGI-58 lacking the putative catalytic seryl residue showed a similar effect on PG levels but not in other phospholipids. These observations led us to test the hypothesis that CGI-58 has a general lipid metabolic function and that it interacts with PXA1.

We have used the yeast two-hybrid system pGADT7-AD (Clontech) and the nuclear relocalization assay vector pRTL2/NLS-RFP (Dhanoa et al., 2010) to construct a yeast clone that expresses GFP-tagged plant CGI-58. This clone was subcloned into the pMDC43 binary vector for two-hybrid interaction with a PXA1 cDNA fragment.

Coimmunoprecipitation of the full-length recombinant proteins in vitro confirmed a physical interaction and mutagenesis studies indicated that the pore-forming domain of PXA1 is a key interacting region of CGI-58. Moreover, in a cgi-58 pxa1 double mutant, plants displayed reductions in jasmonate biosynthesis, auxin signaling, and lipid metabolism consistent with decreased PXA1 activity. We have also provided evidence that recombinant plant CGI-58, but not mouse CGI-58, exhibits LPAAT activity and is able to hydrolyze fatty acid substrates containing an HxxxxD motif. Moreover, a chimera of plsC containing the first 40 amino acids of mouse CGI-58 and an HxxxxD mutation of plant CGI-58 shows copper resistance in BL21(DE3) and a decrease in PG accumulation compared to a control.…
